Motility Control of Neuronal and Human iPS Cells under Serum - and Feeder - Free Culture Condition
نویسنده
چکیده
2 Cell movement is an important step in development. Two experiments were conducted to investigate cell movement control in serumand feeder-free conditions. There were two key findings. 1) The direction of cell migration was affected by the use of a ratchet-shaped micro-chamber. 2) Two proteins were needed as a coating to make the human iPS cell pattern. These findings provide useful tools for a basic study on cell migration and an applied one on regenerative medicine.

The discovery in 2006 that human and mouse fibroblasts could be reprogrammed to generate iPS cells with qualities remarkably similar to embryonic stem cells has created a valuable new source of pluripotent cells for drug discovery, cell therapy, and basic research. GIBCO media and reagents have been at the forefront of pluripotent stem cell research for years. Knockout DMEM supplemented with Kn...
متن کاملA New Feeder-Free Technique to Expand Human Embryonic Stem Cells and Induced Pluripotent Stem Cells
The optimal maintenance of human embryonic stem cells (hESC) in vitro is generally observed in the presence of a feeder-layer of mouse embryonic fibroblasts in a serum-containing medium. Various approaches are now available to remove the feeder requirement. Today, the best feeder-free system for the maintenance of hESC and induced pluripotent stem (iPS) cells is based on a serum-replacement med...

Many investigators have used xenogeneic, especially murine stromal cells and fetal calf serum to maintain and expand human stem cells. The proliferation and expansion of human hematopoietic stem cells in ex vivo culture were examined with the goal of generating a suitable protocol for expanding hematopoietic stem cells for patient transplantation. Using primary fetal liver cells, we established...
متن کاملHuman Induced Pluripotent Stem Cells on Autologous Feeders
BACKGROUND For therapeutic usage of induced Pluripotent Stem (iPS) cells, to accomplish xeno-free culture is critical. Previous reports have shown that human embryonic stem (ES) cells can be maintained in feeder-free condition. However, absence of feeder cells can be a hostile environment for pluripotent cells and often results in karyotype abnormalities. Instead of animal feeders, human fibrob...

BACKGROUND The successful establishment of human induced pluripotent stem cells (hiPSCs) has increased the possible applications of stem cell research in biology and medicine. In particular, hiPSCs are a promising source of cells for regenerative medicine and pharmacology. However, one of the major obstacles to such uses for hiPSCs is the risk of contamination from undefined pathogens in conven...
